Abstract

The synthesis of (3S-8aS)-3-benzyl-1-oxo-hexahydro-2Hpyrrolo(1,2-a) pyrazine was desired for further experimentation on its metabolic effects on day old cockerels. This compound was synthesized using available compounds of L-phenylalanine and L-proline. The intermediate compounds synthesized were L-phenylalanine methyl ester hydrochloride , L-phenylalinol, N-carbobenzoxy1-proline, p-nitrophenyl ester of N-carbobenzoxy-L-proline , N-carbobenzoxy-L-prolyl-L-phenylalainol, N-carbobenzoxy-L-prolylL-phenylalinyl chloride , and L- prolyl-L-phenylalinyl chloride. A discussion of methods of peptide linkage with specific emphasis on the carbobenzoxy N-protecting group and the methods for activating the carboxylic acid of the N-protected amino acid is included. Also discussed are the method and problems encountered in the removal of the carbobenzoxy group from this peptide.
