Abstract
Tef (Eragrostis tef Zucc.) Trotter is a significant grain crop in Ethiopia, occupying over 30% of the country's total cultivated land. Tef is known for its long shelf life, drought resistance, natural pest resistance, versatility in food, beverages, and cattle feed. The crop has recently gained popularity in the United States for its nutritional qualities. Tef has a high content of minerals, amino acids, and is also gluten-free. In the context of climate change and the need for sustainable agriculture, exploring alternative crops to ensure food security is crucial. This research focuses on identifying tef accessions that are well-adapted to the cold climatic conditions of Iowa. To help achieve this objective, we planted 154 tef accessions each on three separate days. We genotyped 28 accessions using short-tandem repeat (STR) markers to differentiate accessions. By multiplex PCR, a total of nine specific loci were amplified using three primers labeled with FAM, Hex, or NED fluorescent dyes. The PCR products were separated by gel electrophoresis to evaluate the performance of each primer pair. Subsequently, fragment analysis was performed by mixing the PCR product with GS600 LIZ standard and Hi-DI formamide. The data collected on allele sizes for STR loci revealed the presence or absence of those specific genetic loci. These findings demonstrate the genetic variation among different tef accessions, offering valuable insights to identify each accession genetically.
