Abstract
The restoration of damaged ecosystems, particularly at superfund sites is a complex issue. These sites present problems with revegetation attempts due to their unique chemical, physical, and biological properties. Revegetation of damaged ecosystems may be enhanced by mycorrhizal associations. The establishment of a facultative mycotroph, Festuca arundinacea Screb. (tall fescue) treated with a commercial fungal inoculum, was studied at a Galena, Kansas mining site. The evaluation of this revegetation project required the identification of endomycorrhizal fungi, Glomus c/arum and G/omus etunicatum in the inoculum. Traditional identification of these fungi based on spore morphology and other physical characteristics is not suitable for differentiating the fungal species found in infected roots. The random amplification of polymorphic DNA (RAPD) has proven useful for developing specific DNA primers to identify fungi under laboratory conditions. However, the development of specific DNA primers for fungal identification in field samples has not been tested. In this study, DNA primers were synthesized and evaluated for use in identifying the presence of G. c/arum and G. etunicatum in root samples of tall fescue from the Galena, Kansas superfund site. The primers 5' GTA AAC CCT AAC CTT AAC AAT CC and 5' AT GAT CTG GTC CTA CAA ACA TTC appear to generate a unique size band for G. clarum, and primers 5' C TGC CGC CAC TAC CAT CAA TAA AAG and 5' C TGC CGC CAC GAG GAA AAT AAT TTG appear to be species specific for G. etunicatum.
